Western blot analysis was performed on whole cell extracts (30 µg lysate) of Hep G2 (Lane 1), A375 (Lane 2), HL-60 (Lane 3) U-937 (Lane 4) and K-562 (Lane 5). The blot was probed with Anti-MAT2B Recombinant Rabbit Monoclonal Antibody (Product # 703221, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 1:5000 dilution). A ~38 kDa band corresponding to MAT2B was observed across the cell lines tested.
Knockdown of MAT2B was achieved by transfecting Hep G2 cells with validated MAT2B specific siRNA (Silencer® select Product # s26227). Western blot analysis (Fig a) was performed using whole cell extracts from the Hep G2 knockdown cells (Lane 3), non-specific scrambled siRNA transfected cells (Lane 2) and untransfected cells (Lane 1). The blot was probed with Anti-MAT2B Recombinant Rabbit Monoclonal Antibody (Product # 703221, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 1:5000 dilution). Densitometric analysis of this Western blot is shown in histogram (Fig b). Loss of signal upon siRNA mediated knock down confirms that antibody is specific to MAT2B.
